Materials and Methods

Preparation of Papaya Leaf Ethanol Extract

Papaya leaf ethanol extract was obtained through the process of maceration using 96% ethanol solvents. The total phenolic content and total flavonoids of this extract were tested before being formulated into three variations of nanogel, namely F1 (5%), F2 (7.5%), and F3 (10%).

Characterization of Nanogel Preparations

Each formula was tested for physical characteristics, stability, antibacterial activity, irritation, and the ability to heal wounds on mouse skin. The results showed that nanogel containing ethanol extract of papaya leaves had a thick consistency, green, homogeneous, stable for 12 weeks at room temperature and low and high temperatures, and did not cause irritation to the rabbit skin.

Results

Antibacterial Activity Test

The antibacterial activity test showed that nanogel containing papaya leaf extract had an inhibition zone diameter of 12.9 mm against Staphylococcus aureus and 13.4 mm against Pseudomonas aeruginosa. These results showed a significant difference compared to negative control groups.

Wound Healing Activity Test

In a group of mice that received 10% papaya leaf extract nanogel, wound healing activity showed a percentage of wound width reduction that healed faster on the 16th day compared to 10% papaya leaf extract gel preparations that showed results on the 18th day. On the 15th day, a group of mice treated with Bioplacenton showed a statistically significant difference compared to other mouse groups. All groups of mice given nanogel and gel showed significant differences compared to the control group that healed on the 21st day.

Histopathological Analysis

Histopathologically, a change in epiderm in nanogel with an average thickness of 78.29 μm compared to 61.98 μm in the gel, as well as the density of collagen in nanogels was more dense than in gels.

Discussion

The results of this study confirmed that the formulation of nanogel ethanol extract of papaya leaves was stable for 12 weeks at room temperature and low and high temperatures. Nanogel with a concentration of 5% papaya leaf extract, 7.5%, and 10% showed the efficacy of better wound healing compared to gels. Nanogel formulation with 10% papaya leaf extract showed the best effective healing of burns in mice.

By understanding the potential of papaya leaf extract in the treatment of burns, this study opens opportunities for the development of nanogel-based topical preparations that are more effective and safe. Utilization of natural ingredients such as papaya leaves not only provides alternatives in wound care but can also reduce the use of antibiotics that have the potential to cause resistance.
